One of the many meanings of en is to describe the use of means of transport.

  en = by
Il va à Paris en voiture.
    He goes by car to Paris.
  Il va aux États-Unis en avion.
    He goes to the US by plane.

But, a bicycle is obviously not something, where a person can be 'in' the means of transport.

  going by bike
Il va en ville à bicyclette.
    He goes by bike to town.
